چکیده Consuming a high-fat and high-calorie diet with a decrease in physical activity causes obesity. Uncoupling protein 1 (UCP-1) is a mitochondrial protein that plays a preventive role against weight gain, obesity and type 2 diabetes. Mitogen-activated protein kinase p38 (MAPK p38) is also involved in processes such as inflammation, obesity, apoptosis and cell differentiation. The aim of the present study was to investigate the effect of aerobic exercise on the expression of UCP-1 and p38MAPK genes in subcutaneous fat tissue in male Wistar rats fed a high-fat diet. In this study, 15 male Wistar rats were randomly divided into three groups: normal control, high-fat control, and high-fat diet + aerobic exercise. After consuming a high-fat diet for 13 weeks, the aerobic exercise group performed the exercise protocol on a treadmill for 12 weeks and 5 sessions per week. The expression of UCP-1 and MAPK p38 genes was measured using RT-PCR method. One-way analysis of variance and Kruskal-Wallis test were used at the P≤0.05 level. UCP-1 gene expression was not significantly different between two groups of normal control and high-fat control (p=0.151); However, a significant difference was observed between the normal control group and the high-fat diet + aerobic exercise group (p=0.008). Also, p38 MAPK gene expression in the high-fat diet control group showed a significant increase compared to the normal control group (p=0.008). While the expression of the p38 MAPK gene in the high-fat diet + aerobic exercise group had a significant decrease compared to the high-fat control group (p=0.008). Aerobic exercise can be a suitable stimulus in UCP-1 mRNA expression. The complexity of p38 pathway activity is caused by its dependence on the type of stimulus and cell.
